This is a combination regimen used in clinical trials as a control arm, consisting of a placebo administered alongside the chemotherapy regimen known as modified FOLFOX6 (mFOLFOX6). The mFOLFOX6 regimen includes three drugs: - Oxaliplatin (a platinum-based chemotherapeutic) - Leucovorin (folinic acid) - 5-Fluorouracil (5-FU) The "placebo" component is an inert substance given to maintain blinding in randomized controlled trials. The active chemotherapy agents work by interfering with DNA synthesis and function in rapidly dividing cells. Oxaliplatin forms DNA crosslinks; 5-fluorouracil inhibits thymidylate synthase and incorporates into RNA/DNA; leucovorin enhances the binding of 5-FU to its target enzyme. This combination is standard for colorectal cancer and sometimes other gastrointestinal cancers[1][2][4][5][6].
