Placenta gp96 is an immunotherapy candidate consisting of the heat shock protein gp96 (also known as HSP90B1 or endoplasmin) purified from human placental tissue. Developed by the Yantai Affiliated Hospital of Binzhou Medical University, it functions as a therapeutic vaccine that leverages the role of gp96 as a molecular chaperone. gp96 naturally binds a broad spectrum of oncofetal antigens, which are proteins expressed during fetal development that are often re-expressed by tumor cells. Upon administration, the gp96-peptide complexes are internalized by antigen-presenting cells (APCs) through the CD91 receptor (LRP1) and activate the APCs via Toll-like receptors 2 and 4. This process facilitates the cross-presentation of tumor-associated antigens to CD8+ T cells, stimulating a targeted anti-tumor immune response. It is currently being investigated in Phase 1 clinical trials for the treatment of advanced solid tumors, including gastric and lung cancers.
