pLADD (personalized Live Attenuated Double-Deleted *Listeria monocytogenes*) is an individualized, neoantigen-based immunotherapy developed by Aduro Biotech for the treatment of microsatellite stable (MSS) metastatic colorectal cancer. Utilizing Aduro's proprietary LADD platform, the therapy consists of attenuated *Listeria monocytogenes* strains engineered to express multiple patient-specific tumor neoantigens identified through genomic sequencing of the patient's tumor. Upon intravenous administration, the *Listeria* vector is taken up by dendritic cells, where it triggers both innate and adaptive immune responses, specifically activating CD8+ T cells against the tumor-specific mutations. Although it reached Phase 1 clinical testing, the program was deprioritized by Aduro in 2019 during a strategic pipeline refocusing.
