The plasmid DNA HIV-1 vaccine (specifically developed as HIVIS-DNA) is an investigational multigene, multiclade DNA-based vaccine candidate designed to prevent HIV-1 infection. Developed under the HIV Vaccine Immunogenicity Study (HIVIS) program by the Swedish Institute for Infectious Disease Control (SMI) in collaboration with the Karolinska Institutet and Tanzanian partners, the vaccine consists of plasmids encoding HIV-1 genes env (subtypes A, B, C), rev (subtype B), gag (subtypes A, B), and RT (subtype B). It is typically evaluated as a priming agent in a heterologous prime-boost regimen, followed by a boost with the recombinant Modified Vaccinia Ankara (MVA-CMDR) vaccine. The vaccine aims to elicit broad cellular and humoral immune responses against multiple HIV-1 clades.
