Overview

The Plasmodium falciparum mosquito challenge is a standardized experimental procedure used to induce malaria infection in healthy volunteers under controlled conditions. It involves the bites of *Anopheles* mosquitoes infected with *Plasmodium falciparum* sporozoites, typically the NF54 strain. This model, extensively utilized and refined by Radboud University Medical Center, serves as a critical tool for evaluating the efficacy of malaria vaccine candidates and prophylactic drugs. In the context of the 'CPS' (Chloroquine Prophylaxis and Sporozoites) immunization strategy, volunteers are exposed to these infected mosquito bites while under chloroquine prophylaxis. This allows the parasite to complete its liver-stage development but prevents symptomatic blood-stage infection, thereby inducing potent, long-lasting protective immunity against subsequent challenges.
