Plasmodium falciparum NF54 sporozoites refer to a whole-organism malaria vaccine approach, specifically utilizing the NF54 strain, developed and extensively studied by Radboud University Medical Center. The primary methodology, known as Chemoprophylaxis and Sporozoites (CPS) immunization, involves exposing healthy volunteers to the bites of mosquitoes infected with NF54 sporozoites while they are concurrently taking antimalarial drugs such as chloroquine or mefloquine. This regimen allows the parasites to reach the liver and initiate early development, which induces a potent and protective immune response against pre-erythrocytic (liver-stage) antigens. The antimalarial drug cover prevents the development of symptomatic blood-stage infection. The NF54 strain, a well-characterized West African isolate, serves as a benchmark for evaluating vaccine efficacy and heterologous protection against other strains in controlled human malaria infection (CHMI) trials.
