Overview

Platelet-coupled IL-2 (P-IL-2) is an experimental cell-based immunotherapy developed by Zhejiang University for the treatment of advanced solid tumors. It consists of the cytokine interleukin-2 (IL-2) coupled to the surface of platelets. This approach leverages the natural tumor-homing and inflammatory-site-targeting properties of platelets to deliver IL-2 directly into the tumor microenvironment. By localizing the delivery of IL-2, P-IL-2 aims to stimulate the proliferation and anti-tumor activity of T cells and natural killer (NK) cells within the tumor while reducing the systemic toxicities, such as vascular leak syndrome, typically associated with high-dose systemic IL-2 therapy. It is currently being investigated in Phase 1 clinical trials as both a monotherapy and in combination with anti-PD-1 checkpoint inhibitors for various malignancies, including esophageal, bladder, liver, ovarian, and small cell lung cancers.
