Overview

Platelet-derived exosome product (PEP) is an exosome-based therapeutic developed by Rion Inc, consisting of extracellular vesicles derived from human platelets. PEP is designed to leverage the regenerative and anti-inflammatory properties of platelet-derived factors. In preclinical models of chronic obstructive pulmonary disease (COPD) and emphysema, nebulized PEP has demonstrated the ability to attenuate lung inflammation and promote tissue homeostasis. Its mechanism involves modulating the lung microenvironment by downregulating pro-inflammatory signaling pathways, such as Thy1-ITGAX/ITGB2, and upregulating reparative pathways like Lamc1-CD44. This orchestration induces a phenotypic switch in alveolar macrophages from a pro-inflammatory state to a reparative M2 state, thereby suppressing the production of tumor necrosis factor (TNF) and other inflammatory mediators. PEP is also being investigated for its potential in wound healing and other regenerative medicine applications.
