Platinum pyrithione is a **synthetic platinum complex** where platinum is coordinated with pyrithione ligands. Unlike traditional platinum-based chemotherapeutics such as cisplatin, platinum pyrithione acts through **strong, selective inhibition of proteasome-associated deubiquitinases (USP14 and UCHL5)** rather than by inducing DNA damage. This results in potent and selective cytotoxicity against a variety of cancer cell lines (including those from leukemia and ovarian cancer), with reduced toxicity in non-cancerous cells and fewer side effects in animal models. Platinum pyrithione demonstrates G2 cell cycle arrest and apoptosis in cancer cells and suppresses tumor growth in xenograft models. It is considered a promising candidate for antineoplastic therapy, particularly for tumors resistant to standard platinum drugs[1][5].
