Overview

Platonin is a **cyanine photosensitizing dye** with notable **antioxidant, anti-inflammatory, immunomodulating,** and **antiplatelet** properties. It has been used to treat trauma, ulcers, and acute inflammation, and has shown neuroprotective effects in preclinical models of ischemic stroke, as well as beneficial effects in animal models of sepsis, heatstroke, ischemia-reperfusion injury, and transplant survival. Mechanistically, platonin acts as a free radical scavenger, suppresses the production of inflammatory cytokines (including interleukin-1β, IL-6, tumor necrosis factor-α, and inducible nitric oxide synthase) via inhibition of transcription factors like nuclear factor-kB (NF-κB) and activator protein-1 (AP-1), and inhibits signaling pathways such as mitogen-activated protein kinases (MAPKs: ERK, JNK, p38 MAPK). Platonin also inhibits platelet aggregation through inactivation of JNK phosphorylation in platelets. Clinically, platonin has been reported for use in immune diseases such as juvenile rheumatoid arthritis and polyarteritis nodosa, mainly in certain Asian countries[1][2].
