PLB-002 is an antibody-drug conjugate (ADC) targeting Claudin 6 (CLDN6), a tight junction protein highly expressed in various solid tumors but with limited expression in normal adult tissues. Developed by PrimeLink BioTherapeutics, the drug consists of a novel humanized anti-CLDN6 single-domain antibody (PLB-002-ab7) conjugated to the microtubule inhibitor eribulin via a proprietary enzyme-cleavable linker, with an average drug-to-antibody ratio (DAR) of 4.0. PLB-002 is designed to deliver the cytotoxic payload specifically to CLDN6-expressing tumor cells, where it is internalized and released to inhibit microtubule dynamics, leading to cell cycle arrest and apoptosis. It is currently being evaluated in Phase 1 clinical trials for advanced solid tumors, including platinum-resistant ovarian cancer, non-small cell lung cancer, and testicular cancer.
