PLB-015

**PLB-015** is a preclinical, HER2-targeted dual-payload antibody-drug conjugate developed by PrimeLink Bio for HER2-positive solid tumors. It uses an anti-HER2 antibody to selectively deliver a topoisomerase I inhibitor and an ATR inhibitor to tumor cells, aiming to combine DNA damage induction with inhibition of the ATR-mediated DNA-damage response and thereby address resistance to topoisomerase I inhibitor payloads. In preclinical HER2-positive cell-line and patient-derived xenograft models, PLB-015 showed potent and durable antitumor activity, favorable pharmacokinetics, and tolerability in cynomolgus monkeys.
