PLD-103 is a discovery-stage small molecule inhibitor being developed by Pelemed to target the YAP-TEAD transcriptional program. The interaction between Yes-associated protein (YAP) and Transcriptional Enhanced Associate Domain (TEAD) transcription factors is the terminal step of the Hippo signaling pathway, which plays a critical role in regulating organ size, cell proliferation, and survival. In many cancers, this pathway is dysregulated, leading to constitutive YAP-TEAD activity that drives tumor growth, metastasis, and resistance to therapy. PLD-103 is designed to disrupt this transcriptional complex, offering a potential therapeutic approach for a variety of YAP-TEAD-dependent malignancies.
