PLD1-selective inhibitors are a class of small molecule therapeutic agents designed to specifically inhibit the enzymatic activity of Phospholipase D1 (PLD1). PLD1 is a lipid-modifying enzyme that hydrolyzes phosphatidylcholine to produce phosphatidic acid (PA), a key second messenger involved in oncogenic signaling pathways such as mTOR and MAPK. In various malignancies, including cervical squamous cell carcinoma (SCC), PLD1 is overexpressed and acts as a critical regulator of cancer metabolism. It promotes glycolytic flux, energy homeostasis, and adaptation to metabolic stress conditions like hypoxia and nutrient deprivation. By selectively blocking PLD1, these inhibitors disrupt the metabolic survival mechanisms of tumor cells, leading to reduced ATP production and increased apoptosis. Research indicates that PLD1 inhibition may be particularly effective in tumor subtypes that exhibit a strong dependency on PLD1-mediated metabolic regulation, such as cervical SCC.
