Overview

AJG522 (Plenvu) is a low-volume (1-liter) polyethylene glycol (PEG)-based osmotic laxative used for bowel cleansing prior to colonoscopy. Developed by Norgine and licensed to EA Pharma and Mochida Pharmaceutical in Japan, it consists of PEG 3350, sodium sulfate, sodium chloride, potassium chloride, and a high dose of ascorbic acid and sodium ascorbate. The mechanism of action involves the osmotic effect of PEG and sulfate, supplemented by the osmotic action of unabsorbed ascorbate in the large intestine, which induces a watery stool and clears the colon. Compared to traditional 2-liter PEG solutions (such as Moviprep/MGV-5), AJG522 offers a reduced fluid intake requirement, which is intended to improve patient compliance and the quality of bowel preparation. It was approved in Japan in March 2024.
