This is a **combination therapy** comprising three agents: **plerixafor** (a selective CXCR4 antagonist), **rituximab** (a monoclonal antibody targeting CD20), and **G-CSF** (granulocyte-colony stimulating factor, a myeloid growth factor). - **Plerixafor** is used to mobilize hematopoietic stem cells (HSCs) from bone marrow to peripheral blood by antagonizing CXCR4, disrupting retention signals mediated by CXCL12, and leading to rapid HSC mobilization. - **Rituximab** is a chimeric monoclonal antibody targeting CD20, expressed on B-lymphocytes, inducing B cell lysis via complement-dependent cytotoxicity (CDC), antibody-dependent cellular cytotoxicity (ADCC), and direct apoptosis. - **G-CSF** stimulates the production and release of neutrophils and progenitor cells from the bone marrow into the bloodstream. When combined with plerixafor, it further enhances stem cell mobilization, particularly for autologous transplantation in patients with lymphoma or multiple myeloma. This combination is studied and sometimes used in the setting of hematopoietic stem cell mobilization for transplant, especially when patients are poor mobilizers or have lymphoid malignancies refractory to standard mobilization regimens, or where it is desired to combine stem cell mobilization with anti-lymphoma therapy[1][2][3][4][5][6].
