Pleurotus ostreatus ethanolic extract (PoEE) is a natural extract derived from the edible oyster mushroom (*Pleurotus ostreatus*). It is primarily investigated in preclinical models for its chemopreventive, antineoplastic, and metabolic properties. PoEE has been shown to exert potent anticancer effects, particularly in hormone receptor-positive breast cancer, by modulating the PI3K/Akt/mTOR signaling pathway. It downregulates key oncogenic components such as PI3K, Akt, mTOR, Ras, and MAPK, while restoring tumor suppressors like PTEN, GSK3β, and FOXO. Additionally, PoEE exhibits anti-obesity potential by suppressing PPARγ expression and maintaining PPARγ promoter methylation during adipocyte differentiation, as well as protecting cells from glycemic and oxidative stress.
