This is a combination nanovaccine technology for cancer immunotherapy, designed for DNA vaccine delivery. It combines poly(lactic-co-glycolic acid) (PLGA) and polyethylenimine (PEI) as nanoparticle carriers co-loaded with two plasmid DNAs encoding programmed death-ligand 1 (pPD-L1) and Dickkopf-1 (pDKK-1). PLGA provides biocompatibility and controlled release, while PEI facilitates gene delivery via endosomal escape. The vaccine aims to induce robust antitumor immune responses by enhancing CD8+ T cell activation—DKK-1 acts as a tumor antigen and pPD-L1 enables immune checkpoint blockade—resulting in increased immunogenicity and tumor growth inhibition. This combination has been studied primarily for multiple myeloma and other hematologic malignancies in preclinical settings[1][4].
