PlGF-2123-144-anti-CD40 is an engineered, extracellular matrix (ECM)-anchored agonistic antibody conjugate developed for localized cancer immunotherapy. It consists of an agonistic anti-CD40 antibody conjugated to a super-affinity peptide derived from placenta growth factor-2 (PlGF-2123-144). This modification enables the construct to bind tightly to the ECM at the site of peritumoral injection, resulting in prolonged tissue retention and significantly reduced systemic exposure. By anchoring locally, the drug minimizes systemic side effects, such as liver toxicity and cytokine release, which are common with unmodified anti-CD40 therapies. Its mechanism of action involves the activation of CD40 on antigen-presenting cells, such as dendritic cells and B cells, which in turn stimulates a robust T-cell-mediated antitumor immune response. Preclinical studies have demonstrated enhanced efficacy in various tumor models, including those resistant to checkpoint inhibitors.
