Overview

PlGF-2123-144-anti-CTLA4 is an experimental biologic conjugate designed for localized cancer immunotherapy. It consists of a monoclonal antibody targeting the immune checkpoint protein cytotoxic T-lymphocyte-associated protein 4 (CTLA4) covalently linked to a highly heparin-binding peptide fragment (residues 123–144) derived from placenta growth factor-2 (PlGF-2). This specific PlGF-2 sequence serves as an extracellular matrix (ECM)-binding domain, exhibiting exceptionally high affinity for various ECM components including laminin, fibronectin, and collagen. When administered peritumorally or intratumorally, the PlGF-2 domain anchors the antibody to the local tumor microenvironment, preventing its rapid systemic dissemination. This localized retention is intended to maintain high therapeutic concentrations within the tumor to drive antitumor T-cell activity while minimizing systemic exposure, thereby reducing the risk of immune-related adverse events (irAEs) such as liver toxicity and systemic cytokine release. Preclinical studies in melanoma models have demonstrated that this approach preserves the efficacy of CTLA4 blockade while significantly improving the safety profile compared to systemic administration of the unmodified antibody.
