PLK-1 UsiRNA-DiLA2 is a preclinical-stage RNA interference (RNAi) therapeutic developed by Marina Biotech (formerly MDRNA, Inc.). The drug consists of a UsiRNA (unlocked nucleic acid-modified siRNA) designed to target and silence the mRNA of polo-like kinase 1 (PLK1), a critical regulator of the cell cycle (specifically the G2/M transition and cytokinesis). The UsiRNA is delivered using Marina Biotech's proprietary DiLA2 (Di-alkylated Amino Acid) lipid-based liposomal delivery system. In preclinical studies, the therapeutic demonstrated significant knockdown of PLK1 expression and tumor growth inhibition in models of bladder cancer, hepatocellular carcinoma, and malignant ascites associated with ovarian and gastrointestinal cancers.
