PLK1 siRNA is a small interfering RNA specifically designed to target the mRNA of polo-like kinase 1 (PLK1), a serine/threonine kinase that is a key regulator of cell cycle progression, particularly at the G2/M transition. PLK1 is highly expressed in many types of cancer and is considered essential for proliferation and survival of rapidly dividing tumor cells. Delivery of PLK1 siRNA leads to sequence-specific cleavage and silencing of PLK1 mRNA via the RNA interference (RNAi) pathway, resulting in reduced PLK1 protein levels. This induces mitotic arrest and apoptosis selectively in cancer cells, and demonstrates potent inhibition of tumor cell proliferation and tumor growth in multiple cancer models, including non-small cell lung cancer, triple-negative breast cancer, and pediatric B-cell leukemia. Various delivery platforms for PLK1 siRNA have been investigated, including nanoparticles and lipid-based formulations, to enhance systemic delivery and tumor targeting[1][3][4][6][7].
