PLL-g-PEG is a cationic graft copolymer consisting of a poly(L-lysine) backbone with polyethylene glycol side chains. Calm Water Therapeutics develops its proprietary PLL-g-PEG material as **PegPlus**, a preservative-free topical ophthalmic polymer intended to improve tear-film integrity and ocular-surface barrier function in dry eye disease. In a completed 28-day controlled study, a 2% PLL-g-PEG ophthalmic formulation containing glycerin and PEG 400 improved dry-eye signs and symptoms versus a marketed lubricant-eye-drop control. Preclinical studies also found that PLL-g-PEG inhibited macropinocytic uptake of the antibody-drug conjugate rituximab-mc monomethyl auristatin F by human corneal epithelial cells, supporting investigation for prevention of antibody-drug-conjugate-associated corneal toxicity. PLL-g-PEG itself is not an approved standalone drug or an INN-designated active ingredient.
