PLN-101325

PLN-101325 is a monoclonal antibody developed by Pliant Therapeutics for the treatment of muscular dystrophies, including Duchenne muscular dystrophy. It acts as an allosteric agonist of the integrin α7β1 (composed of ITGA7 and ITGB1 subunits), a receptor upregulated in muscle cells as a compensatory mechanism in various forms of muscular dystrophy. By activating this receptor, PLN-101325 aims to stabilize muscle cell membranes, increase muscle strength, reduce muscle damage, and improve overall muscle function. Preclinical studies have shown that treatment with PLN-101325 improves cell morphology, tissue organization, diaphragm strength, and respiratory function in animal models and human cells affected by muscular dystrophy.
