PLN-1013416 (also known as P1213416) is an orally bioavailable, potent, and highly selective small molecule inhibitor of the **αvβ1 integrin**, developed by **Pliant Therapeutics**. The αvβ1 integrin is primarily expressed on activated myofibroblasts and plays a critical role in the activation of latent **transforming growth factor-beta (TGF-β)**, a master regulator of fibrosis. By selectively targeting αvβ1, PLN-1013416 aims to inhibit TGF-β activation locally in fibrotic tissues while avoiding the systemic toxicities associated with broad TGF-β inhibition. The drug is being developed primarily for the treatment of **Primary Sclerosing Cholangitis (PSC)**, a chronic liver disease characterized by progressive bile duct fibrosis. It has completed Phase 1 clinical evaluation in healthy volunteers to assess safety, tolerability, and pharmacokinetics.
