PLN-1177 is a small molecule pan RGD (Arg-Gly-Asp) integrin inhibitor developed by Pliant Therapeutics for the treatment of fibrotic liver diseases, including primary sclerosing cholangitis (PSC). It was designed to target the RGD-binding site of multiple integrins, which are known to play a role in the activation of TGF-beta and other pro-fibrotic pathways. While preclinical studies showed that PLN-1177 could reduce serum total bilirubin and inhibit biliary fibrosis, further research indicated that non-selective pan RGD inhibition might exacerbate inflammation in cholestatic liver disease. Consequently, Pliant Therapeutics shifted its focus toward more selective integrin inhibitors, such as PLN-1561 and PLN-1705, and did not advance PLN-1177 into clinical development.
