Overview

PLN-1474 is an orally available, small molecule selective inhibitor of the integrin αvβ1. It was originally discovered and developed by Pliant Therapeutics for the treatment of nonalcoholic steatohepatitis (NASH) with liver fibrosis and other fibrotic liver diseases. The drug acts by selectively blocking αvβ1 integrin-mediated activation of transforming growth factor-beta (TGF-β), a key driver in the development of fibrosis. Preclinical studies demonstrated that PLN-1474 reduced liver fibrosis in animal models. After successful completion of Phase 1 clinical trials showing favorable safety and pharmacokinetic profiles, development rights were transferred to Novartis under a collaboration agreement; however, Novartis later discontinued its clinical development in NASH and returned global rights to Pliant Therapeutics[2][3][5][7][10].
