Overview

PLN-1561 is an investigational, orally bioavailable small molecule inhibitor developed by Pliant Therapeutics that selectively targets the integrin alpha-V beta-1 (αvβ1). This integrin is a critical mediator in the localized activation of transforming growth factor-beta (TGF-β), a potent pro-fibrotic cytokine that drives the progression of scarring in various organs. By specifically inhibiting αvβ1, PLN-1561 is designed to block TGF-β activation directly within fibrotic tissues, potentially treating conditions such as Primary Sclerosing Cholangitis (PSC) and other forms of liver fibrosis. This targeted approach aims to provide therapeutic efficacy while avoiding the systemic toxicities and safety concerns associated with broad, non-selective TGF-β inhibition. The drug is intended to reduce collagen deposition and biliary scarring, thereby improving liver function and slowing disease progression in patients with chronic cholestatic liver diseases.
