Overview

PLN-1705 is an investigational, orally bioavailable, highly selective small molecule inhibitor of integrin αvβ1, developed by Pliant Therapeutics. It is primarily being evaluated for the treatment of primary sclerosing cholangitis (PSC) and other liver-related fibrotic diseases. Integrin αvβ1 is a key mediator in the activation of transforming growth factor-beta (TGF-β), a master regulator of fibrosis, specifically within activated myofibroblasts in the liver. By selectively inhibiting the αvβ1 integrin, PLN-1705 aims to block the localized activation of TGF-β, thereby reducing the progression of biliary fibrosis and collagen deposition. This targeted approach is intended to provide potent anti-fibrotic activity while minimizing the systemic side effects often associated with non-selective TGF-β inhibition.
