PLN-75068 is an investigational, orally bioavailable small molecule selective inhibitor of the αvβ1 integrin, developed by Pliant Therapeutics. It is primarily being developed for the treatment of liver fibrosis, including conditions such as primary sclerosing cholangitis (PSC). The αvβ1 integrin is highly expressed on activated fibroblasts (myofibroblasts) and plays a critical role in the local activation of transforming growth factor-beta (TGF-β), a key driver of fibrotic tissue deposition. By selectively targeting the αvβ1 isoform, PLN-75068 aims to inhibit TGF-β activation specifically within the fibrotic microenvironment, potentially offering a safer profile than non-selective TGF-β inhibitors.
