PLN-76104 is an investigational, orally bioavailable, small molecule selective inhibitor of the αvβ1 integrin, developed by Pliant Therapeutics. The αvβ1 integrin is a critical mediator of TGF-β activation, a central driver of fibrosis in various organs, particularly the liver. By specifically targeting αvβ1, PLN-76104 aims to inhibit the localized activation of TGF-β within fibrotic tissue while avoiding the systemic toxicities associated with broad TGF-β inhibition. The drug is primarily being developed for the treatment of primary sclerosing cholangitis (PSC), a chronic cholestatic liver disease characterized by progressive biliary fibrosis and inflammation.
