Overview

Plocabulin (PM-060184) is a marine-derived small molecule tubulin polymerization inhibitor being developed by PharmaMar. It is a microtubule-destabilizing agent that binds to a unique site on the β-tubulin subunit, acting as an interfacial inhibitor to reduce microtubule dynamicity. Originally isolated from the Mediterranean sponge *Lithistida*, the compound has demonstrated potent antitumor activity in various solid tumors. It is administered intravenously and has been investigated in Phase II clinical trials for the treatment of advanced colorectal cancer and hormone receptor-positive, HER2-negative breast cancer, both as a monotherapy and in combination with other chemotherapeutic agents like gemcitabine.
