Plonmarlimab (TJ003234) is a first-in-class, humanized immunoglobulin G1 (IgG1) monoclonal antibody that targets and neutralizes granulocyte-macrophage colony-stimulating factor (GM-CSF). By inhibiting GM-CSF signaling, it aims to reduce inflammation and cytokine release. It was developed by I-Mab Biopharma for the treatment of diseases characterized by excessive or dysregulated immune activation such as cytokine release syndrome (CRS), severe COVID-19, macrophage activation syndrome, hemophagocytic lymphohistiocytosis, and rheumatoid arthritis. The drug has reached Phase 3 clinical trials for COVID-19-related CRS and Phase 2 for other hyperinflammatory conditions[4][5][6].
