Overview

Plozalizumab (formerly MLN1202 or TAK-202) is a humanized monoclonal antibody that acts as an antagonist of the C-C chemokine receptor type 2 (CCR2). CCR2 is a chemokine receptor expressed on monocytes, macrophages, and certain T-cell subsets, and its activation by ligands such as CCL2 (MCP-1) is a key driver of monocyte recruitment to sites of chronic inflammation and the tumor microenvironment. By binding to CCR2, plozalizumab prevents the recruitment and activation of these inflammatory cells. Developed by Millennium Pharmaceuticals (a subsidiary of Takeda), the drug has been investigated for a variety of inflammatory and autoimmune conditions, including rheumatoid arthritis, multiple sclerosis, and atherosclerotic cardiovascular disease, as well as for diabetic nephropathy. In oncology, it has been evaluated for its potential to modulate the immune environment in bone metastases and in combination with checkpoint inhibitors for advanced melanoma.
