**Plozalizumab + nivolumab** is an experimental combination therapy evaluated for advanced melanoma. **Plozalizumab** is a monoclonal antibody targeting CCR2, aiming to modulate the tumor microenvironment by blocking chemokine-driven recruitment of immunosuppressive cells. **Nivolumab** is a monoclonal antibody that blocks the programmed death-1 (PD-1) receptor on T cells, restoring anti-tumor immune responses. This combination was assessed in a phase 1b clinical trial to determine if targeting both CCR2-mediated immunosuppression and PD-1–dependent T cell exhaustion could enhance anti-tumor efficacy beyond PD-1 inhibition alone. In early results, the combination did not show sufficient efficacy to warrant further enrollment in advanced melanoma[1].
