Drug intelligence / Profile preview
PLT011 (formerly EI-011) is a preclinical humanized bifunctional fusion protein consisting of an anti-PD-L1 monoclonal antibody linked to interleukin-10 (IL-10) polypeptides. Developed by Pilatus Biosciences (originally discovered by Elixiron Immunotherapeutics), PLT011 is designed to target the tumor microenvironment by binding to PD-L1, thereby blocking the immunosuppressive PD-1/PD-L1 pathway. Simultaneously, the localized IL-10 component stimulates the IL-10 receptor on tumor-infiltrating CD8+ T cells, promoting metabolic reprogramming (oxidative phosphorylation) and rejuvenating exhausted T cells. This dual mechanism aims to enhance anti-tumor immunity with reduced systemic toxicity, showing promising efficacy in preclinical models of hepatocellular carcinoma (HCC) and melanoma.
