Overview

PluraliNK cells are a universal, "plug-and-play" natural killer (NK) cell therapy platform engineered to be redirected by various monoclonal antibodies (mAbs) and T-cell engagers (TCEs). Developed by researchers at MD Anderson Cancer Center, these cells are modified using polycistronic vectors to express the full CD3 complex, an invariant TCR α/β (iTCR), high-affinity CD16A (F158V), and IL-15 for cytokine support. This engineering allows the cells to be activated through both CD3/TCR- and Fc-dependent pathways, enabling synergistic combination with existing immunotherapies to target multiple antigens and overcome tumor heterogeneity or immune escape. The platform is designed to provide a safer, more flexible alternative to traditional CAR-NK cells by requiring external antibody-mediated signals for activation, and it has demonstrated potent cytotoxicity in both hematologic and solid tumor models.
