PLX138 is a long-acting C-type natriuretic peptide (CNP) analog being developed by ProLynx for the treatment of achondroplasia. The therapeutic utilizes ProLynx's proprietary drug delivery technology, which involves a hydrogel-based system with a self-cleaving linker to provide sustained release of the active CNP peptide. This formulation is designed to overcome the extremely short half-life of native CNP, potentially allowing for weekly or less frequent subcutaneous administration. By acting as an agonist for the natriuretic peptide receptor B (NPR-B), PLX138 inhibits the overactive fibroblast growth factor receptor 3 (FGFR3) signaling pathway, which is the primary driver of impaired bone growth in achondroplasia patients.
