PlyF307SQ-8C is an engineered, recombinant bacteriophage-derived endolysin (lysin) developed by ContraFect Corporation, specifically designed to target and kill *Acinetobacter baumannii*. It is a truncated and optimized variant of the native PlyF307 lysin, which was originally isolated from an *A. baumannii* phage. The molecule acts as a peptidoglycan hydrolase, rapidly degrading the bacterial cell wall upon contact, which leads to osmotic lysis and cell death. PlyF307SQ-8C has shown significant potency against multidrug-resistant (MDR) and carbapenem-resistant *A. baumannii* (CRAB) strains in preclinical studies, including the ability to disrupt biofilms. While it served as a key lead in ContraFect's Gram-negative program, the company's clinical focus shifted to CF-370 (a related engineered lysin) before ContraFect ceased operations and entered liquidation in late 2023.
