Overview

PM-4321 is an orally available small-molecule antagonist of the aryl hydrocarbon receptor (AHR) being developed as an antineoplastic agent. By inhibiting AHR signaling, PM-4321 is designed to counteract tumor-promoting and immune-suppressive pathways driven by endogenous and tumor-derived AHR ligands, thereby enhancing antitumor immunity and directly impairing cancer cell survival. Preclinical studies have shown dose-dependent tumor growth inhibition in syngeneic colorectal cancer models, supporting its potential as a targeted immunomodulatory therapy in oncology.[1][16]
