Overview

PM-mb15-41BBL is an experimental cell therapy candidate and technology platform developed by ProMab Biotechnologies, specifically engineered to enhance the therapeutic efficacy of natural killer (NK) cells. The construct incorporates two key signaling molecules: membrane-bound Interleukin-15 (mbIL-15) and 4-1BB ligand (4-1BBL, also known as CD137L). The mbIL-15 component provides continuous, localized pro-survival and proliferative signals through the IL-15 receptor complex, mimicking natural trans-presentation and reducing the requirement for systemic cytokine administration. Simultaneously, 4-1BBL provides potent co-stimulatory signals via the 4-1BB (CD137) receptor, which is critical for the activation, metabolic fitness, and cytotoxic function of NK cells. While widely utilized as a specialized feeder cell line (K562-based) for the ex vivo expansion of clinical-grade NK cells, PM-mb15-41BBL is also being developed as a direct therapeutic modality to improve the persistence and anti-tumor activity of adoptive cell transfers in the treatment of solid tumors and hematologic malignancies.
