PM0503

PM0503 is a small molecule 4-alkanoylaminobenzamide developed by Pharmacyl AB for the treatment of ulcerative colitis (UC). It acts as an inhibitor of NFκB activation by specifically preventing the degradation of IκBβ, an inhibitory protein that sequesters NFκB in the cytoplasm. By stabilizing IκBβ, PM0503 prevents the translocation of NFκB to the nucleus, thereby reducing the transcription of pro-inflammatory genes. Preclinical studies in dextran sulphate sodium (DSS)-induced colitis models have shown that PM0503 significantly reduces the disease activity index (DAI). Furthermore, PM0503 has demonstrated synergistic effects when combined with 5-aminosalicylic acid (5-ASA) agents such as sulfasalazine, which typically target the breakdown of IκBα, providing a rationale for combination therapy in mild-to-moderate UC.
