Overview

PM1009 is a fully human bispecific antibody that targets both TIGIT and PVRIG immune checkpoint receptors. It consists of an anti-TIGIT IgG1 fused with an anti-PVRIG single-chain variable fragment (scFv) at the C terminus. By simultaneously blocking the TIGIT/CD155/CD112 and PVRIG/CD112 signaling pathways, PM1009 abrogates inhibitory signals that suppress T cell and natural killer (NK) cell activation in the tumor microenvironment. This dual blockade enhances cytotoxic T lymphocyte (CTL) and NK cell activity, promotes immune-mediated tumor cell killing, inhibits tumor proliferation, and supports depletion of TIGIT-positive regulatory T cells via FcγR engagement. Developed originally by Biotheus (now acquired by BioNTech), PM1009 is being investigated primarily for advanced solid tumors and liver cancer in early-phase clinical trials[1][2][3][4][5].
