Overview

PM1016 (also known as TILT-123) is an oncolytic adenovirus biologic developed through a partnership between Biotheus and Tilt Biotherapeutics. It is a chimeric Ad5/3 adenovirus engineered for tumor-selective replication via a 24-base pair deletion in the E1A region and the inclusion of an E2F promoter. The virus serves as a vector for two potent immune-stimulating cytokines: human Tumor Necrosis Factor-alpha (TNF-α) and human Interleukin-2 (IL-2). By inducing tumor cell lysis and locally secreting these cytokines, PM1016 aims to enhance T-cell infiltration and activation within the tumor microenvironment, potentially overcoming resistance to other immunotherapies like checkpoint inhibitors. While it has been evaluated in Phase I/II clinical trials for advanced solid tumors, the development under the PM1016 code name was recently terminated.
