PM1022 is a recombinant, fully human bispecific antibody that targets both PD-L1 (programmed death-ligand 1) and TIGIT (T cell immunoglobulin and ITIM domain). It is engineered as an IgG1 wild type Fc backbone with a VHH at the C-terminus targeting PD-L1. The mechanism of action involves dual blockade of the PD-L1/PD-1 and CD155/CD112/TIGIT pathways, aiming to release T cell inhibition, deplete regulatory T cells in the tumor microenvironment, and engage Fcγ receptors to activate myeloid cells. PM1022 has demonstrated robust preclinical efficacy in vitro and in vivo. It is currently being evaluated as monotherapy for advanced solid tumors in Phase I/II clinical trials[1][3][4][7].
