Overview

PM1032 is a humanized bispecific antibody that targets both claudin 18.2 (CLDN18.2) and tumor necrosis factor receptor superfamily member 9 (4-1BB). It acts as a conditional agonist of 4-1BB in the tumor microenvironment by crosslinking via CLDN18.2 expressed on tumor cells, leading to T-cell and natural killer (NK) cell activation specifically at the site of CLDN18.2-positive tumors[1][5][7]. This mechanism results in potent anti-tumor immune responses with limited systemic toxicity, as immune activation is restricted to areas where CLDN18.2 is present[1][5]. The drug is being developed primarily for advanced or metastatic solid tumors, especially gastrointestinal cancers such as gastric cancer that express CLDN18.2[3][7]. Clinical trials are ongoing to evaluate its safety and efficacy in these indications[3].
