PM21 particles are nano-scale plasma membrane vesicles derived from a modified K562 cell line (K562-mb21-41BBL) engineered to express 4-1BB ligand (4-1BBL) and membrane-bound interleukin-21 (mbIL-21). These particles serve as a feeder cell-free platform for the robust ex vivo and in vivo expansion and activation of natural killer (NK) cells for cancer immunotherapy. By providing localized co-stimulatory signals, PM21 particles induce high levels of NK cell proliferation and enhance their cytotoxic activity against various malignancies, including acute myeloid leukemia (AML) and solid tumors like ovarian cancer. The technology was developed by researchers at the University of Central Florida and Nationwide Children's Hospital, utilizing cell lines provided by Kiadis Pharma (a Sanofi company).
