Overview

PMB-101 is an autologous chimeric antigen receptor T cell (CAR‑T) therapy that targets the B‑cell activating factor receptor (BAFF‑R/CD268) on malignant B cells, being developed by PeproMene Bio in collaboration with City of Hope for the treatment of relapsed or refractory B‑cell acute lymphoblastic leukemia (B‑ALL) and other B‑cell malignancies.[7][15] The CAR construct is based on humanized anti‑BAFF‑R monoclonal antibody–derived scFv domains and a second‑generation signaling backbone, enabling potent and selective cytotoxicity against BAFF‑R–expressing lymphoid tumors, including those that have lost CD19 expression after prior CD19‑directed therapies.[7] In preclinical CD19‑resistant lymphoma and leukemia models, BAFF‑R CAR‑T cells induced marked tumor regression and prolonged survival, supporting clinical development as a first‑in‑class BAFF‑R–targeted cell therapy for patients who have failed existing B‑cell–directed immunotherapies.[7][9]
