Drug intelligence / Profile preview
PMB-102 (also known as PMB-CT01) is an investigational, first-in-class autologous chimeric antigen receptor T-cell (CAR-T) therapy developed by PeproMene Bio. It is designed to target B-cell activating factor receptor (BAFF-R), a member of the tumor necrosis factor (TNF) receptor superfamily that is expressed almost exclusively on B cells and is essential for their survival. PMB-102 is constructed using an anti-BAFF-R single-chain variable fragment (scFv) antibody coupled with second-generation CD3ζ and 4-1BB signaling domains. The therapy is currently being evaluated in a Phase 1 clinical trial (NCT05370430) for the treatment of patients with relapsed or refractory B-cell non-Hodgkin lymphoma (r/r B-NHL), including mantle cell lymphoma (MCL), follicular lymphoma (FL), and diffuse large B-cell lymphoma (DLBCL). Because BAFF-R is required for B-cell survival, targeting this receptor is expected to prevent tumor escape via antigen downregulation, offering a promising alternative for patients who have relapsed after or are ineligible for CD19-targeted CAR-T therapies.
