PMB-107 is a monoclonal antibody developed by Protheragen that specifically targets the Glycoprotein-A Repetitions Predominant (GARP)/Latent-associated peptide (LAP)-TGFβ1 complex. GARP is a transmembrane protein primarily expressed on the surface of activated regulatory T cells (Tregs) and platelets, where it functions to anchor and present latent TGF-β1. The activation and subsequent release of TGF-β1 from this complex is a key mechanism of immune evasion in the tumor microenvironment, contributing to the suppression of anti-tumor T-cell responses. By binding to the GARP/LAP-TGFβ1 complex, PMB-107 prevents the liberation of active TGF-β1, thereby reversing Treg-mediated immunosuppression and potentially enhancing the efficacy of other immunotherapies. PMB-107 is currently in Phase I clinical development for the treatment of solid tumors.
